Basically during a blower door test you figure out how leaky a home is.
This is done by depressurizing the house using the door cover and fan. Then a device called a manometer measures the airflow through the fan. After doing some math, based on the dimensions of the house, we can figure out how much air leaks from your home.
The bottom line is it's required by building code for all new homes. The point is to ensure that the craftsmanship on the home was done to a certain standard, and since air leaking isn't something residents would notice right away, it gets tested right after its built.
